Abstract
Seed oil percentage is not the only criterion to evaluate the improvement of sesame oil. The value of a breeding material is determined by the fatty-acid composition, the content of lignans, the content of tocopherols, the production of seeds, as well as their stability when grown under various conditions. These qualities are linked and an improvement in one does not necessarily lead to a similar improvement in the others. The review focuses on the use of genetically and molecular evidences reported in sesame for a more coherent approach to oil quality improvement. It explores how metabolism, variation (natural and induced), associations with the genome, and techniques for selection/modification of desired traits are related. This review focuses especially on evidence for individual targets. A gene identified by developmental expression or association analysis has a lesser level of support than a gene examined by biochemical characterisation, allelic analysis or experimental manipulation. The studies of fatty acid desaturation offer a practical link between sequence variation and oil composition while the studies of sesame lignans provide examples of well-defined enzymatic functions. New technologies such as mapping, transcriptomics, lipidomics and small-RNA studies have expanded the list of candidates to be explored. However, their value in breeding will rely on how those candidates are validated and put into appropriate genetic backgrounds. The evidence supports a coordinated approach integrating conventional breeding, mutagenesis, molecular selection and biotechnology, reflecting the guidance from biochemical phenotyping and evaluation across environments. The development of the materials should be assessed by what can be done with them, not by just the number of molecular discoveries.
